To determine the mediation of the motor action of ghrelin, corticotropin releasing hormone CRH ; antagonist, the dopamine antagonist haloperidol, the nitric oxide synthase NOS ; inhibitor Nnitro-L-arginine-methyl ester L-NAME ; or the serotonin 5-HT 2 ; antagonist cgproheptadine were administered to the rats. The motor responses were diminished by the preadministration of the CRH antagonist and haloperidol, while L-NAME and cyproheptadind did not prove to be effective in reducing the ghrelin-evoked behavioural response in the open-field test. Further haloperidol, being administered 30 min before the peptide treatment, transiently and significantly reduced spontaneous locomotor activity. Present data suggest, that in the behavioural responses evoked by ghrelin, both CRH release and the action of the dopaminergic neurons of the subcortical motor system might be involved. Cachexia is late stage outcome of cancer anorexia, occurs in 80%-90% of patients with advanced cancer, and includes wasting of body protein and fat, with concomitant loss of appetite. Malnutrition of cancer patients is a severe clinical problem due to increased mortality and morbidity. There is no clincal treatment available to cure the cachectic syndrome, but increasing knowledge on mechanisms eliciting the cachectic response in severe disease may improve targeted pharmaceutical intervention to restore cachexia. As an example, antiinflammatory treatment has been successfully used to reduce tumor growth and improve cachexia. It has also been observed that cancer patients treated with antidepressants seem to increase food intake and gain weight. These findings may be due to alterations in the neurotransmitter serotonin 5HT ; system, known to play an important role in appetite regulation. The aim of this study was to evaluate whether alterations in the serotonin system influenced development of cancer cachexia in an experimental tumor model in mice . Material and method: Forty C57Bl 6J mice were implanted with a methylcholanthrene-induced sarcoma bilaterally in the flanks. This experimental tumor model in mice induce a severe anorexia-cachexia syndrome as previously described. An additional 24 non tumor bearing mice were investigated as healthy controls for 19 days. Ten mice per group were injected intraperitoneally with: a ; saline control group ; b ; Citalopram a selective serotonin reuptake inhibitor blocking the synaptic uptake of 5HT 20mg kg day from day 1to 9. c ; Cyproheptadine, a 5 HT receptor antagonist, 5mg kg day from day 1 to 9. Parachlorophenylalanine PCPA ; , a serotonin synthesis inhibitor, 300 mg kg day from day 6 to 9. Food intake and body weight were recorded daily. Net tumor growth was assesed over 10 days. Plasma concentrations of IL-6 and PGE2, and body composition were analyzed. Results: No acute toxic reactions were observed following 19 days of treatment in healthy control mice. In tumorbearing mice, treatment with citalopram, cyproheptadine or PCPA did not alter food intake, nor was cachexia improved. Tumor growth was not altered. Conclusion: Alterations in serotoninergic system by citalopram, cyproheptadine or PCPA treatment did not attenuate development of cancer cachexia in this experimental tumor model. Our results suggest that cancer cachexia, at least in this model, operates through mechanisms other than the serotoninergic system.
